Irisin is a recently described exercise-induced hormone secreted by skeletal muscle in mice and humans. Irisin activates beige fat cells (beige cells have a gene expression pattern distinct from either white or brown fat and are preferentially sensitive to the polypeptide hormone irisin). Irisin is cleaved from the type I membrane protein FNDC5 and improves systemic metabolism by increasing energy expenditure. FNDC4 is an ortholog of FNDC5 with 50% identity and 86% similarity compared to Irisin. FNDC4 as well as FNDC5 are extremely well conserved between species. The function of FNDC4 is unknown. The human FNDC4 gene is highly enriched in liver, brain tissue and adipocytes.

Members of this family are chromatin-associated and widely spread in the nucleus of higher eukaryotic cells. HMGB2 contains 2 HMG box DNA-binding domains. It is associated with chromatin and has the ability to bend DNA, preferentially single-stranded DNA. It is shown that HMGB2 is able to efficiently bend DNA and form DNA circles. In addition, HMGB2 is involved in the final ligation step in DNA end-joining processes of DNA double-strand breaks repair and V(D)J recombination.

The primary structure contains 28 cysteine residues forming multiple disulfide bonds. DBP acts as a multifunctional protein found in plasma, ascitic fluid, cerebrospinal fluid, and urine and on the surface of many cell types. DBP binds to vitamin D and its plasma metabolites and transports them to target tissues. Glial Cell Line-Derived Neurotrophic Factor (GDNF) is a disulfide-linked homodimeric glycoprotein that belongs to the TGF- beta superfamily. It has been shown to promote the survival of various neuronal subpopulations in both the central as well as the peripheral nervous systems at different stages of their development. Human GDNF cDNA encodes a 211 amino acid residue prepropeptide that is processed to yield a dimeric protein. Mature human GDNF was predicted to contain two 134 amino acid residue subunits. Cells known to express GDNF include Sertoli cells, type 1 astrocytes, Schwann cells, neurons, pinealocytes and skeletal muscle cells. Human Brain-Specific Angiogenesis Inhibitor 3 (BAI3) is a 177 kDa seven-span transmembrane (TM) protein, which is thought to be a member of the secretin receptor family. It is synthesized by neurons of the CNS and likely is a negative regulator of angiogenesis. BAI3 is 1498 amino acids in size. It contains three distinct regions: an N-terminal extracellular domain (ECD) (aa25-883), a 7-TM segment, and a C-terminal cytoplasmic region. The ECD contains four antiangiogenic TSP type 1 repeat (aa296-508), and one GSP domain (aa 816-867) that is likely used to cleave the ECD from the membrane-bound receptor. There is one altermate splice form that shows a deletion of aa 643-665. Over aa 25-880, human BAI3 shares 98% aa identity with mouse BAI3. BAI3 has been reported primarily in the brain, but is also localized to lung, testis, and pancreas. It might be involved in angiogenesis inhibition and suppression of glioblastoma.
