Peroxiredoxin-4 (PRDX4) is a member of the AhpC/TSA family. PRDX4 is a cytoplasmic protein and contains one thioredoxin domain. PRDX4 exists in homodimer or heterodimer with PRDX1. PRDX4 reduces hydrogen peroxide and alkyl hydroperoxides to water and alcohol with the use of reducing equivalents derived from thiol-containing donor molecules. In addition, PRDX4 is probably involved in redox regulation of the cell, regulating the activation of NF-kappa-B in the cytosol by a modulation of I-kappa-B-alpha phosphorylation.

CD4 is accessory protein for MHC class-II antigen/T-cell receptor interaction. CD4 induces the aggregation of lipid rafts. CD4 is a primary receptor used by HIV-1 to gain entry into host T cells. CD84, also called SLAMF5, is a member of the CD2 subgroup of the immunoglobulin receptor superfamily. Members of this CD2 subgroup mediate signal transduction through the interaction of its immunoreceptor tyrosine-based switch motifs (ITSM) in the intracellular region and the SH2 domain of adaptor molecules SAP (SLAM-associated protein) and EAT-2 (EWS-activated transcript 2), and accordingly modulate both adaptive and innate immune responses. CD84 expression has been documented on several hematopoietic cell types, including monocytes, macrophages, dendritic cells, B lymphocytes, and platelets. Activation of cell surface CD84 initiates a signallling cascade involving its intra-cytoplasmic tyrosine residues that results in Bcl-2 upregulation, which in turn enhances cell survival. Either immunoneutralisation or blockade of CD84 with a CD84 extracellular domain protein fragment induces cell death in vitro and in vivo.

Insulin-like growth factors (IGFs) comprise a family of endocrine, paracrine and autocrine polypeptides consisting of the ligands IGF1 and IGF2, two receptors (IGF1R, IGF2R), at least 6 IGF-binding proteins (IGFBPs) and IGFBP proteases. Among the binding proteins, IGFBP6 is unique because of its N-terminal disulphide linkages and its marked binding preference for IGF2. It is a potent inhibitor of the interaction between IGF2 and its receptor IGF1R, thus preventing major functions of IGF2, such as induction of proliferation, differentiation, cell adhesion, or colony formation. In particular, IGFBP-6 inhibited the growth of neuroblastoma and rhabdomyosarcoma xenografts. GFBP-6 is expressed in many tissues, including lung, liver, gut and the central nervous system.

Fetuin-B is a member of the Fetuin family that is part of the Cystatin superfamily of Cysteine Protease inhibitors. It is reported that Fetuin-B is highly expressed in liver tissue, in tongue and placenta tissues. Fetuin-B is a paralogue of Fetuin-A. Fetuin-A and Fetuin-B display similarities and differences in their characteristics, however, they share only 20% amino acid sequence identity. The amounts of Fetuin-B in human serum, unlike Fetuin-A, vary with gender and are higher in females than in males. Fetuin-B is an inhibitor of basic calcium phosphate precipitation but is less active than Fetuin-A. Fetuin-B expression is decreased in Fetuin-A deficient knock-out mice. The expression of Fetuin-B has been shown to be regulated by FXR (Farnesoid X Receptor), a nuclear receptor activated by bile acids. Evidence has shown that overexpression of Fetuin-B in skin squamous carcinoma cells suppresses tumor growth in nude mice. The function of Fetuin B is still not fully characterized.
