Serine Hydroxymethyltransferase Cytosolic (SHMT1) is a member of the SHMT family. SHMT1 is a cytoplasmic protein and exists as a homotetramer. SHMT1 catalyzes the reversible conversion of serine and tetrahydrofolate to glycine and 5,10-methylene tetrahydrofolate. This reaction provides one carbon unit for the synthesis of methionine, thymidylate, and purines in the cytoplasm. A reduction in SHMT1 levels would result in less glycine that could affect the nervous system by acting as an agonist to the NMDA receptor and this could be a mechanism behind Smith-Magenis syndrome.

IL-36Ra/IL-1F5 is a highly and a specific antagonist of the IL-1 receptor-related protein 2-mediated response to IL-36alpha (IL-1F6), IL-36beta (IL-1F8) and IL-36gamma (IL-1F9). These cytokines bind to IL-36R (IL-1Rrp2) and IL-1RAcP, activating similar intracellular signals as IL-1. IL-36Ra inhibits the production of proinflammatory cytokines, including IL-12, IL-1beta, IL-6, TNF-alpha and IL-23 induced by IL-36 in BMDC and CD4 T cells. Skin and dendritic cells are target of the IL-36 interleukins leading to a Th1 response. Recently mutations that affect the levels and the activity of IL-36Ra have been found in patients with pustular psoriasis, leading to enhanced production of inflammatory cytokines (IL-8 in particular) by keratinocytes.

Insulin-growth factor-like gene family is a new family of proteins consisting of four proteins in humans (IGFL1 to 4) and one in mice (mIGFL). mIGFL is expressed in normal skin in mice and further upregulated during inflammation responses in skin or after skin wounding. In human only IGFL1 expression is increased in psoriatic skin samples. mIGFL and human IGFL1 and 3 interact with specificity and high affinity to a novel receptor named IGF-like family receptor 1 (formerly TMEM-149). TIM-4 is exclusively expressed on antigen-presenting cells, where it mediates phagocytosis of apoptotic cells and plays an important role in maintaining tolerance. TIM-4 binds specifically to TIM-1 which is also the cellular receptor for the hepatitis A virus, and has been implicated in the development of asthma. Among hematopoietic cells, TIM-1 is expressed on activated B and T cells, preferentially in the Th2 subset of CD4+ T cells. The interaction of TIM-4 with TIM-1 induces costimulatory and hyperproliferative signals in T cells.

It complexes with IFN- alpha/ beta R1 to form the signaling receptor complex for the family of alpha and beta IFN subtypes. By alternative splicing, IFN- alpha/ beta R2 can exist as a secreted soluble protein or as a type I membrane protein. IFN- alpha/ beta R2 is the principal ligand binding subunit of the receptor. Ligand binding is stabilized by the subsequent association with IFN- alpha/ beta R1, resulting in the formation of a signaling ternary receptor complex. IFNAR2 was detected in most lymphocytes, monocytes, and granulocytes, although IFNAR2 expression was higher in the monocytes and granulocytes than in the lymphocytes. Among the lymphocyte subsets, IFNAR2 showed high expression in natural killer (NK) cells and low expression in T lymphocytes. Isoform 1 and isoform 3 of IFNAR2 are directly involved in signal transduction due to their interaction with the TYR kinase, JAK1. Isoform 1 also interacts with the transcriptional factors, STAT1 and STAT2. Both forms are potent inhibitors of type I IFN activity.
