Bone Morphogenetic Protein Receptor II (BMPR-II) is a Type II Serine/Threonine Kinase that mediates cellular responses to BMPs. BMPR-II is characterized by lacking of a GS domain, and presence of a C-terminal extension typical of type II receptors. Mouse interleukin 13 (mIL-13) is a pleiotropic cytokine produced by activated Th2 cells. IL-13 induces B cell proliferation and immunoglobin production. It contains a four helical bundle with two internal disulfide bonds. Mouse IL13 shares 58% sequence identity with human protein and exhibits cross-species activity. IL13 signals via receptor IL13R (type2, IL4R) and activates STAT-6. IL13 initially binds IL-13R alpha1 with low affinity and triggers association of IL4R alpha, generating a high affinity heterodimeric receptor IL13R and eliciting downstream signals. IL13 also binds IL-13R alpha2 with high affinity, which plays a role in a negative feedback system of IL13 signaling. IL13 is an important mediator of allergic inflammation and disease.

Interleukin-33 (IL-33) was initially discovered as a nuclear factor NF-HEV abundantly expressed in high endothelial venules. It is a 30-32 kD pro-inflammatory protein with intracellular and extracellular activities and a chromatin-associated cytokine of the IL-1 family with high sequence and structural similarity to IL-1 and IL-18. IL-33 is highly and selectively expressed by high endothelial venule endothelial cells (HEVECs) in human tonsils, Peyers's patches, and lymph nodes. It contains a bipartite nuclear localization signal at the C-terminus, and is targeted to the nucleus when ectopically expressed in human umbilical vein endothelial cells (HUVECs) and HeLa cells. The C-terminal fragment, corresponding to mature IL-33, binds and triggers signaling. IL-33 mediates its biological effects via Toll-interleukin 1 (IL-1) receptor (TIR) domain-containing receptor ST2, activates NF-kappaB and MAP kinases, and drives production of T(H)2-associated cytokines from in vitro polarized T(H)2 cells. In vivo, IL-33 induces the expression of IL-4, IL-5, and IL-13 and leads to severe pathological changes in mucosal organs. Human IL-33 is 270 amino acids in length.

IL6R is a protein complex consisting of a IL-6 receptor subunit (IL6R) and interleukin 6 signal transducer Glycoprotein 130. IL6R also denotes the human gene encoding this subunit. Alternatively spliced transcript variants encoding distinct isoforms have been reported. IL6R subunit also shared by many other cytokines. The soluble form of IL6R arises from proteolytic cleavage of membrane-bound IL6Rα, and acts agonistically by making the IL6 ligand accessible to the signal transducer gp130. Dysregulated production of IL6 and IL6R are implicated in the pathogenesis of several inflammatory diseases and malignancies such as multiple myeloma, rheumatoid arthritis, or osteoporosis, and it has been reported that a humanised anti-IL6R monoclonal antibody is a promising agent applicable to the therapeutic approach for IL6 driven diseases.
