SERPINB1 is a member of the serpin family and Ov-serpin subfamily. As protease inhibitors, serpins have an array of functions including regulating blood coagulation, fibrinolysis, the complement pathway, angiogenesis, inflammation, tumour suppression, extracellular matrix remodelling, and cell motility. SERPINB1 regulates the activity of the neutrophil proteases elastase, cathepsin G, proteinase-3, chymase, chymotrypsin, and kallikrein-3. Reactive bond 1 of SerpinB1 is specific for reaction with chymotrypsin-like protease such as cathepsin G, chymotrypsin or chymase. Reactive bond 2 of SerpinB1 is specific for reaction with elastase-like protease such as neutrophyl elastase, proteinase-3, pancreatic elastase or PSA. In addition, SERPINB1 also functions as a potent intracellular inhibitor of granzyme H.

SERPINI2 is a secreted protein of the serpin family. Serpins are a group of proteins with similar structures that were first identified as a set of proteins able to inhibit proteases. As protease inhibitors, serpins have an array of functions including regulating blood coagulation, fibrinolysis, the complement pathway, angiogenesis, inflammation, tumour suppression, extracellular matrix remodelling, and cell motility. SERPINI2 is expressed in human tissues including pancreas and adipose tissues. Mutations of human SERPINI2 can directly result in conditions such as acinar cell apoptosis and malabsorption.

Human B7-H2 is a member of the growing B7 family of immune costimulatory proteins. Other family members include B7-1, B7-2, B7-H1 (PD-L1), PD-L2 and B7-H3. B7 proteins are members of the immunoglobulin (Ig) superfamily, the extracellular domains contain 2 Ig-like domains and all members have short cytoplasmic domains. B7-H2 has been identified as the ligand for ICOS, a member of the CD28 family of costimulatory receptors. Human B7-H2 is expressed constitutively on resting B cells, dendritic cells and at low levels on monocytes. The B7-H2/ICOS interaction appears to play roles in T cell dependent B cell activation and Th differentiation.

Interleukin-2 (IL-2) is a 133 amino acid glycoprotein with one intramolecular disulfide bond and variable glycosylation. It is secreted by activated T cells and induces proliferation and maturation of activated T cells, natural killer cells, and lymphokine activated killer cells. IL-2 also stimulates proliferation of antibody-producing B cells, activates neutrophils, and induces mononuclear cells to secrete IFN-gamma and TNF-alpha and -beta. Moreover, studies have shown that IL-2 is required for activation-induced apoptosis, an important hemeostatic mechanism in the immune system, which is involved in the maintenance of peripheral tolerance to self-antigens.
