Glycogen synthase kinase 3 (GSK3) is a serine/threonine kinase that is involved in the regulation of many signaling pathways. To date, 2 isoforms have been identified: GSK3alpha and GSK3beta. Specifically, GSK3beta has been shown to play a key inhibitory role in both the insulin and Wnt signaling pathways (Papkoff and Aikawa 1998). Gap junctional intercellular communication is thought to play a key role in development and may also be involved in epilepsy (Aronica et al., 2001). Connexin 43 forms gap-junctional channels and regulates the permeability of these gap junctions to small organic molecules. Permeability of connexin 43 is known to be regulated by phosphorylation at Ser368 by protein kinase C (Yogo et al., 2002; Bao et al., 2004a). Phosphorylation of Ser368 by PKC induces a conformational change of connexin 43 that results in a decrease in gap junction permeability (Bao et al., 2004b).

Although differing in substrate specificity, subcellular localization, and tissue distribution, all isozymes of this family convert free long-chain fatty acids into fatty acyl-CoA esters, and thereby play a key role in lipid biosynthesis and fatty acid degradation. This isozyme is highly expressed in brain, and preferentially utilizes myristate, arachidonate, and eicosapentaenoate as substrates.
