Serine Protease Inhibitor Kazal-Type 4 (SPINK4) is a secreted protein containing one Kazal-like domain. SPINK4 is a member of the SPINK protein family. The gene family of serine protease inhibitors of the Kazal type (SPINK) are functional and positional candidate genes for celiac disease (CD). SPINK1 plays an important role in protecting the pancreas against excessive trypsinogen activation. It is a potent natural inhibitor of pancreatic trypsin activity. SPINK1 mutations are associated with the development of acute and chronic pancreatitis and have been detected in all forms of chronic pancreatitis. SPINK2 functions as a trypsin/acrosin inhibitor and is synthesized mainly in the testis and seminal vesicle where its activity is engaged in fertility. The SPINK2 protein contains a typical Kazal domain composed by six cysteine residues forming three disulfide bridges. SPINK9 was identified in human skin. Its expression was strong in palmar epidermis, but not detectable or very low in non palmoplantar skin.

Interleukin-6 (IL-6) is a multifunctional factor, which exerts its activities through binding to a highaffinity receptor complex consisting of two membrane glycoproteins: an 80 kDa component receptor that binds IL-6 with low affinity (IL-6Ralpha) and a signal transducing component of 130 kDa (gp130) that does not bind IL-6 by itself, but is required for highaffinity binding of IL-6 by the complex. A soluble form of the IL-6Ralpha has been found in the urine of healthy adult humans.

Interleukin-22 (IL-22), also known as IL-10 related T cell derived inducible factor (ILTIF) was initially identified as a gene induced by IL-9 in mouse T cells and mast cells. IL-22 has been shown to activate STAT1 and STAT3 in several hepatoma cell lines and upregulate the production of acute phase proteins. IL-22 is produced by normal T cells upon anti-CD3 stimulation in humans. Mouse IL-22 expression is also induced in various organs upon lipopolysaccharide injection, suggesting that IL-22 may be involved in inflammatory responses. The functional IL-22 receptor complex consists of two receptor subunits, IL-22R(CRF29) and IL-10Rbeta(CRF24), belonging to the class II cytokine receptor family.

Description:
Interleukin-18 (IL-18) is a costimulatory factor for production of interferon-gamma (IFN-gamma) in response to toxic shock and shares functional similarities with IL-12. IL-18 is synthesised as a precursor 24kDa molecule without a signal peptide and must be cleaved to produce an active molecule. IL-1 converting enzyme (ICE; Caspase-1) cleaves pro-IL-18 at aspartic acid in the P1 position, producing the mature, bioactive peptide that is readily released from the cells. It is reported that IL-18 is produced from Kupffer cells, activated macrophages, keratinocytes, intestinal epithelial cells, osteoblasts, adrenal cortex cells and murine diencephalon. IFN-gamma is produced by activated T or NK cells and plays critical roles in the defense against microbiral pathogens. IFN-gamma activates macrophages and enhances NK activity and B cell maturation, proliferation and Ig secretion. IFN-gamma also induces expression of MHC class I and II antigens and inhibits osteoclast activation. IL-18 acts on T helper type-1 (Th1) T cells and in combination with IL-12 strongly induces them to produce IFN-gamma. Pleiotropic effects of IL-18 have also been reported, such as enhancement production of IFN-gamma and GM-CSF in peripheral blood mononuclear cells, production of Th1 cytokines, IL-2, GM-CSF, IFN-gamma in T cells and enhancement of Fas ligand expression by Th1 cells.

Cluster of Differentiation 74 (CD74) is also known as HLA class II histocompatibility antigen gamma chain (HLADG), DHLAG, Ia-GAMMA, invariant chain (Abbreviated Ii). CD74 / HLADG is a polypeptide involved in the formation and transport of MHC class II protein. The cell surface form of the invariant chain is known as CD74. CD74 is upregulated in several cancers and is also expressed by non-immune cells during inflammation. CD74 plays a critical role in MHC class II antigen processing by stabilizing peptide-free class II alpha / beta heterodimers in a complex soon after their synthesis and directing transport of the complex from the endoplasmic reticulum to the endosomal / lysosomal system where the antigen processing and binding of antigenic peptides to MHC class II takes place. CD74 / DHLAG also serves as cell surface receptor for the cytokine MIF.

Chemokine (C C Motif) Ligand 26 (CCL26) is a novel small cytokine belonging to the CC chemokine family, which involved in immunoregulatory and inflammatory processes. CCL26 is expressed constitutively in thymus, but only transiently in phytohemagglutinin-stimulated peripheral blood mononuclear cells. It specifically binds and induces chemotaxis in T cells and elicits its effects by interacting with the chemokine receptor CCR4. Eotaxin-3/CCL26, along with Eotaxin-1 and Eotaxin-2, selectively activates the CC chemokine receptor 3 (CCR3). The Eotaxin-3-CCR3 interaction may play an important role in allergic diseases such as atopic dermatitis and bronchial asthma. The full-length cDNA for Eotaxin-3 encodes a protein of 94 amino acids with a putative signal peptide of either 23 or 26 amino acid residues. Both the 71 and 68 amino acid residue variants of recombinant Eotaxin-3 demonstrate equal potency in inducing chemotaxis of a human CCR3-transfected cell line. Unlike most other CC chemokines, Eotaxin-3 maps to human chromosome 7q11.2, within 40 kilobases of the Eotaxin-2 loci. Eotaxin-3 and Eotaxin-2 are unique in that they are the only chemokines identified to date that map to chromosome 7.

alpha Crystallin B Chain (CRYAB) is a cytoplasmic protein that belongs to the small heat shock protein (HSP20) family. Alpha crystallins are composed of two gene products: alpha-A and alpha-B, for acidic and basic, respectively. Alpha crystallins can be induced by heat shock and are members of the small heat shock protein (sHSP also known as the HSP20) family. Alpha crystallins acts as molecular chaperones and hold them in in large soluble aggregates. CRYAB is expressed widely in many tissues and organs. It may contribute to the transparency and refractive index of the lens. The deficiency of CRYAB is the cause of myopathy myofibrillar type 2 (MFM2) and cataract posterior polar type 2 (CTPP2).

Ephrin Type-B Receptor 1 (EPHB1) is a single-pass type I membrane protein that belongs to the Ephrin-B family of receptor tyrosine kinases that is involved in embryonic nervous and vascular system development. EPHB1/EPHT2 contains two fibronectin type-III domains, one protein kinase domain and one SAM (sterile alpha motif) domain. EPHB1 could stimulate fibroblast motility on extracellular matrix in a kinase-dependent manner, which also correlated with its association with Grb7, an adaptor molecule implicated in the regulation of cell migration. It binds to ephrin-B1, ephrin-B2 and ephrin-B3. EPHB1 plays an important roles in diverse biological processes including nervous system development, angiogenesis, and neural synapsis formation and maturation and may be involved in cell-cell interactions in the nervous system.

S100A8 is a member of the S100 family, EF-hand superfamily of Ca-binding proteins. It is produced by neutrophils and monocytes, and forms Ca2+-dependent heterodimer/heterotetramer complexes (termed calprotectin) with S100A9. Calprotectin (S100A8/A9) which has a wide plethora of intra- and extracellular functions. The intracellular functions include: facilitating leukocyte arachidonic acid trafficking and metabolism, modulation of the tubulin-dependent cytoskeleton during migration of phagocytes and activation of the neutrophilic NADPH-oxidase.

UCHL1 is a component of the ubiquitin system, which has a fundamental role in regulating various biological activities. UCHL1 is a thiol protease that recognizes and hydrolyzes a peptide bond at the C-terminal glycine of ubiquitin. UCHL1 also binds to free monoubiquitin and may prevent its degradation in lysosomes.
