Mesencephalic astrocyte-derived neurotrophic factor(MANF) is a secreted protein which belongs to the ARMET family. MANF selectively promotes the survival of dopaminergic neurons of the ventral mid-brain. It modulates GABAergic transmission to the dopaminergic neurons of the substantia nigra. MANF enhances spontaneous, as well as evoked, GABAergic inhibitory postsynaptic currents in dopaminergic neurons. MANF inhibits cell proliferation and endoplasmic reticulum (ER) stress-induced cell death. The N-terminal region of ARMET may be responsible for neurotrophic activity while the C-terminal region may play a role in the ER stress response. MANF reduces endoplasmic reticulum (ER) stress and has neurotrophic effects on dopaminergic neurons. Intracortical delivery of recombinant MANF protein protects tissue from ischemic brain injury. MANF has been described as a survival factor for dopaminergic neurons. MANF and a homologous protein, the conserved dopamine neurotrophic factor (CDNF), form a novel evolutionary conserved family of neurotrophic factors. MANF expression was widespread in the nervous system and non-neuronal tissues.

Interferon alpha/ beta Receptor 2 (IFN- alpha/ beta R2) is a single-pass type I membrane protein which belongs to the type II cytokine receptor family. It complexes with IFN- alpha/ beta R1 to form the signaling receptor complex for the family of alpha and beta IFN subtypes. By alternative splicing, IFN- alpha/ beta R2 can exist as a secreted soluble protein or as a type I membrane protein. IFN- alpha/ beta R2 is the principal ligand binding subunit of the receptor. Ligand binding is stabilized by the subsequent association with IFN- alpha/ beta R1, resulting in the formation of a signaling ternary receptor complex. IFNAR2 was detected in most lymphocytes, monocytes, and granulocytes, although IFNAR2 expression was higher in the monocytes and granulocytes than in the lymphocytes. Among the lymphocyte subsets, IFNAR2 showed high expression in natural killer (NK) cells and low expression in T lymphocytes. Isoform 1 and isoform 3 of IFNAR2 are directly involved in signal transduction due to their interaction with the TYR kinase, JAK1. Isoform 1 also interacts with the transcriptional factors, STAT1 and STAT2. Both forms are potent inhibitors of type I IFN activity.

AHSG is an important circulating inhibitor of calcification in vivo, and is downregulated during the acute-phase response. Once released in serum, it further polymerises forming dimers to octamers. The degree of serum polymerisation is critical for the biological activity of MBL. Fibroblast growth factor 21 (FGF-21) is a member of the fibroblast growth factor family and has been identified as an important regulator of energy metabolism connecting nutrition, growth, reproduction and longevity. FGF-21 lacks the heparin-binding domain of most FGF proteins, allowing its secretion. FGF-21 is abundantly expressed in adipose tissues, liver and pancreas. In adipose tissue, FGF-21 promotes glucose uptake and oxidation and in liver it replenishes on fasting the tissues with fuel on low nutritional supply. It also induces lipolysis and ketogenesis. FGF-21 acts through FGF receptors (FGFR) associated with the auxiliary protein beta-Klotho. Recently, FGF-21 has been reported to interact directly with the brain circadian clock to coordinate activity and reproduction as part of the adaptation to fasting. Due to its multiple functions of normalizing glucose, lipid and energy homeostasis, FGF-21 represents an attractive novel therapy for type 2 diabetes mellitus and obesity.
