BAP3 Antibody: BAP3 was initially identified through interaction in a yeast two-hybrid system with the brain-specific angiogenesis inhibitor 1, a p53-target gene that encodes a seven-span transmembrane protein member of the secretin receptor family. BAP3 is predominantly expressed in the brain and possess high homology with Munc13 and synaptotagmin, suggesting that BAP3 may play a role in regulating neurotransmitter release. Recent experiments have shown that BAP3 is induced in certain tumors such as desmoplastic small round cell tumor. Ectopic expression of BAP3 in tumor cells dramatically enhances growth in low serum conditions and colony formation in soft agar, suggesting that the regulated exocytotic pathway may play a role in cancer cell proliferation.

RTP801 Antibody: RTP801 was initially identified as a gene induced by DNA damage, and later found to also be regulated by other cellular stresses such as hypoxia and glucocorticoid treatment. Recently, RTP801 has been shown to act as a mediator of tuberous sclerosis complex (TSC)-dependent regulation of the mammalian Target of Rapamycin (mTOR), an evolutionarily conserved serine/threonine kinase that regulates cell growth and cell cycle. In response to energy stress, RTP801 inhibits mTOR function, resulting in dephosphorylation of downstream targets such as ribosomal protein S6 kinase 1 and 4EBP1 and decreasing cell growth. Disregulation of RTP801 may thus contribute to human tumorigenesis.

SAPK/Erk kinase (SEK1), also known as MKK4 or Jun kinase kinase (JNKK), activates the MAP kinase homologues SAPK and JNK in response to various cellular stresses and inflamatory cytokines. Activation of SEK1 occurs through MEKK phosphorylation of serine and threonine residues at positions 257 and 261, respectively. Considerable evidence indicates that a Polo-Like Kinase (PLK) plays an important role in cell cycle regulation. PLK is also required for bipolar spindle formation, activation of the anaphase-promoting complex/cyclosome, and cytokinesis. Recent work led to the identification of a PLKK that is thought to be responsible for activation of PLK. Recent work (Erikson, et al., 2004) has shown that PLKK is in turn activated by phosphorylation at three sites (Ser482, Ser486 and Ser490). Thus activation of PLK is thought to involve a kinase cascade involving the phosphorylation of Ser482,486,490 in PLKK.

In addition to its role in plasticity, the expression of synapsin I is a precise indicator of synapse formation. Thus synapsin I immunocytochemistry provides a valuable tool for the study of synaptogenesis. The role of synapsin in synaptic plasticity and in synaptogensis is regulated by phosphorylation. Serine 9 is the site on synapsin I that is phosphorylated by cAMP-dependent protein kinase and by calcium calmodulin kinase I. Phosphorylation of this site is thought to regulate synaptic vesicle function.
